Calculating Fluorescence Intensity at poles

I was wondering if there is an easy way to calculate the FI at the poles, where the midline is used to partition the cell into left pole, middle, and right pole?

Do you want to partition the fluorescence of the whole cell into three parts; left pole, right pole and middle?
How would be midline be used here? When referring to 'midline' do you mean the line along the long axis of the cell, from the middle of the left pole to the right pole?
