In the debug perspective, specifically in variable view , can the the "Location" column be checked by default

Do you mean in your own product based on CDT, or in CDT's own default.

For the former, I think you can do it by providing a default pref_state_memento.org.eclipse.debug.ui.VariableView preference for org.eclipse.debug.ui using a plugin_customization.ini file via your product definition.

For changing CDT, I don't think we would change that as Location is not valuable for most users AFAIK
